The effect of aerobic exercise and probiotics on PRDX6 gene expression in adipose tissue of obese rats

Background: Obesity, characterized by chronic low-grade inflammation and oxidative stress, is a major risk factor for various metabolic disorders. Peroxiredoxin 6 (PRDX6), an important antioxidant enzyme, plays a key role in maintaining redox homeostasis and modulating inflammatory processes. The present study aimed to evaluate the effects of aerobic exercise and probiotic supplementation—individually and in combination—on PRDX6 gene expression in the visceral adipose tissue of obese male Wistar rats.
Materials and methods: In this experimental design, 40 rats were initially assigned to either a standard diet or a high-fat diet (HFD) enriched with fructose to induce obesity over 24 weeks. Following successful obesity induction, the animals were randomly divided into five subgroups: control, HFD (obese), HFD + exercise, HFD + probiotic, and HFD + exercise + probiotic. The aerobic exercise intervention consisted of treadmill running for 8 weeks, while the probiotic treatment was administered orally during the same period. At the end of the interventions, expression levels of the PRDX6 gene in visceral fat were analyzed using quantitative Real-Time PCR.
Results: HFD significantly increased obesity-related parameters and decreased PRDX6 expression compared to controls (P=0.001). Neither aerobic exercise nor probiotics alone led to a significant increase in PRDX6 expression. However, their combined application significantly upregulated PRDX6 gene expression compared to the obese group (P=0.046).
Conclusion: In conclusion, the combination of aerobic training and probiotic supplementation demonstrates a synergistic effect in enhancing PRDX6 expression and may serve as a promising non-pharmacological strategy for counteracting obesity-related oxidative stress and metabolic dysfunction
