Background: Actinomycetes are among the groups of microorganisms that are widely distributed in nature. The increase in antibiotic resistance has outpaced the discovery of new antibiotics. This study aimed to evaluate the antimicrobial activity of ethyl acetate and methanol extracts of lesser-known actinomycetes from olive orchard soils in Rudbar County, located in the Alborz Mountains. Materials and methods: The antimicrobial activity of two strains EA7 and EA6 identified based on Bergey’s reference book was investigated against the studied microorganisms (Staphylococcus aureus and Pseudomonas aeruginosa) by crossover and well-plate methods in the primary screening. The antimicrobial activity of the fermentation medium, ethyl acetate and methanol extracts of the strains was investigated using sterile paper discs and well-plate methods, respectively. The diameter of the halo was reported in millimeters (mm). Results: Strain EA7 showed greater antimicrobial activity than strain EA6 against the studied microorganisms (Staphylococcus aureus) in primary and secondary screening. Also, the antimicrobial activity of the extracts was greater against gram-positive bacteria than gram-negative bacteria. In comparison, ethyl acetate extract showed greater antimicrobial activity than methanol extract. Conclusion: The difference in the antimicrobial results of the extracts is related to the difference in the cell wall structure of the two groups of bacteria studied (Staphylococcus aureus and Pseudomonas aeruginosa). The results of the study showed that strain EA7, due to its greater antimicrobial effect, can be a potential source for obtaining new bioactive compounds and antibiotics.
