fa بررسی کمی بیان ژن های PALB2 و BRIP1 در جمعیت زنان ایرانی مبتلا به سرطان پستان در مقایسه با گروه شاهد Quantitative investigation of BRIP1 and PALB2 expression level in Iranian women with breast cancer compared to normal samples ژنتيك Genetic تجربي Experimental سابقه و هدف: سرطان پستان شایعترین سرطان در دنیا و دومین علت مرگ و میر پس از سرطان ریه در زنان است. هدف از این تحقیق بررسی کمی بیان ژنهای BRIP1 و PALB2 در نمونه خون افراد مبتلا به سرطان سینه نسبت به افراد شاهد سالم و بررسی نقش این دو ژن به عنوان بیومارکر جهت غربالگری بود. روش بررسی: پژوهش موردی شاهد زیر به بررسی سطح بیان ژنهای PALB2 و BRIP1 در 50 نمونه از افراد بیمار در مقایسه با 50 نمونه شاهد انجام شد. RNA تام استخراج، cDNA سنتز و بیان PALB2 و BRIP1 در حضور ژن کنترل GAPDH با استفاده از روش  qRT-PCR مورد بررسی قرار گرفت. یافتهها: افزایش معنیدار بیان بیومارکرهای هدف در افراد سرطانی نسبت به نرمال مشاهده شد، به طوری که ژنهای BRIP1 و PALB2 به ترتیب 419/3 و 326/7 برابر افزایش بیان نشان دادند (001/0=p). همچنین نتایج با استفاده از فرمول 2-ΔΔCt  با توجه به میانگین Ct افراد سالم  مورد ارزیابی قرار گرفت و grade بیماری با میزان بیان ژنها ارتباط معنیداری را نشان داد، به طوری که با افزایش مرحله و درجه  شدت سرطان بیان بیومارکرها افزایش مییافت (003/0=p). نتیجه گیری: با توجه به  افزایش معنیدار بیان ژنهای BRIP1 و PALB2 در افراد بیمار در مقایسه با افراد نرمال شاهد، احتمالا افزایش بیان سبب مهار فعالیت ژن های سرکوب کننده تومور میشود. با توجه به افزایش بیان این ژنها در نمونههای سرطانی، این ژنها  را میتوان به عنوان نشانگر احتمالی مرتبط با سرطان سینه جهت بررسیهای تکمیلی معرفی کرد. Background: Breast Cancer is the most common cancer in the world and it is the second reason of death between women following the lung cancer. The aim of this study was quantitative evaluation of BRIP1 and PALB2 genes expression level in blood samples of breast cancer comparing with normal people to investigate the role of these two genes as biomarkers during breast cancer diagnosis and screening. Materials and methods: In this case- control study, we investigated the expression level of BRIP1 and PALB2 genes among 50 breast cancer patients in comparison with 50 control cases. Total RNA from blood samples were extracted and cDNA was synthesized. The specific primer for detection of gene markers was designed and expression level of BRIP1 and PALB2 in presence of GAPDH control gene was quantitatively studied by using qRT-PCR. Results: The level of BRIP1 andPALB2 gene expression was significantly increased in breast cancer patients compared to control population. Results showed quantitative increase of BRIP1 and PALB2 genes with 3.419 and 7.326 fold respectively for breast cancer compared to normal samples (P=0.001). Also, the results were evaluated using the formulas 2-ΔΔCt according to the mean Ct of healthy subjects and grading of the disease showed a significant relationship with the expression of genes, so that by increasing staging and severity of cancer, the expression of biomarkers increased (p=0.003). Conclusion: Based on significant increasing expression of PALB2 and BRIP1 genes in breast cancer patients in our study, probably increasing expression of these genes will inhibit the activity of tumor suppressor genes. So, we can introduce these genes as biomarker of breast cancer in the diagnostic work up of disease.
