fa سنتز و تعیین اثر آپوپتوتیک نانوفلورید کلسیم بر روی سلول های سرطانی پستان Synthesis and determination of apoptotic effect of calcium nanofluoride on breast cancer cells سابقه و اهداف: سرطان پستان از بیماریهای شایع زنان در سراسر جهان میباشد و دارای عوامل متعددی است. تنظیم کلسیم در تومور زایی سلول های سرطانی و تکثیر سلولی، مهاجرت، متاستاز و مقاومت آپوپتوز نقش ضروری دارد. نانوکلسیم با تغییر در PH سلول های سرطانی باعث مهار تومور می گردد. روش بررسی: مخلوط کربنات کلسیم با فلورید هیدروژن سونیکه شد و نانوذره با تکنیک های DLS و TEM تایید شد. سپس سلول های سرطانی MCF-7، MDA-MB-231، MDA-MB-468، BT-474 و سلول سالم MSC کشت داده شد. همچنین تست MTT برای تعیین  IC50دارو و انجام ادامه تست های آپوپتوز، چرخه سلولی، خراشیدگی، اسفیرزایی، تهاجم و کلنی زایی انجام شد. یافته ها: میزان IC50 تیمار نانوفلوریدکلسیم با استفاده از تست MTT برابر 2/1 میلی گرم بر میلی لیتر گردید اما برای تست های آپوپتوز و چرخه سلولی از 6/0میلی گرم بر میلی لیتر استفاده شد که میزان آپوپتوز8/2درصد در سلول های ,MDA-MB-23117درصد در سلول های MCF-7و4درصد در سلول های BTبدون میزان معنادار نشان داد. همچنین چرخه سلولی  MDA-MB-231 در مراحل G1و  G2به ترتیب افزایش7/6 و01/1درصد و در مرحله S کاهش79/5  را بدون میزان معناداری را نشان داد.  MCF-7نیز در مراحل G1وG2 به ترتیب کاهش46/11و43/6درصد، در مرحله Sافزایش52/22درصد داشت. BT در مراحل G1و G2 افزایش 64 درصد را نشان می دهد. نتیجه گیری: نانوفلورید کلسیم با راه اندازی آپوپتوز، موجب افزایش مرگ سلولی گردیده، عملکرد خوبی در مهار و کاهش کلنی زایی، بستن محل خراشیدگی، تهاجم، اسفیرزایی سلول های سرطان پستان رده سلولی MDA-MB-231 از خود نشان داد.   ‌Background & objectives:  Breast cancer is one of the most common diseases in women around the world and has many causes. Calcium regulation plays an essential role in cancer cell tumorigenesis and cell proliferation, migration, metastasis and resistance to apoptosis. Nanocalcium inhibits tumor by changing the pH of cancer cells. In this study, calcium nanofluoride was synthesized and evaluated as one of the treatment candidates and reducing symptoms in breast cancer.   Materials & methods: The mixture of calcium carbonate was sonicated with hydrogen fluoride and the nanoparticles were confirmed by DLS and TEM techniques. MCF-7, MDA-MB-231, MDA-MB-468, BT-474 and healthy MSC cancer cells were cultured. MTT test was also performed to determine the IC50 of the drug and to perform tests for apoptosis, cell cycle, abrasion, hemorrhage, invasion and colonization. Results: The IC50 level of calcium nanofluoride treatment was 1.2 mg / ml using MTT test, but 0.6 mg / ml was used for apoptosis and cell cycle tests with an apoptosis rate of 2.8% in MDA cells. MB-23117% in MCF-7 cells and 4% in BT cells showed no significant extent. Also, MDA-MB-231 cell cycle in G1 and G2 stages showed an increase of 6.7% and 1.01%, respectively, and in S stage showed a decrease of 5.79% without significant extent. MCF-7 decreased by 11.46 and 6.43 percent in G1 and G2 stages, respectively, and increased by 22.52 percent in S stage. BT in stages G1 and G2 shows a 64% increase.
