fa بررسی تاثیر سویه‌های حساس و مقاوم مایکوباکتریوم توبرکلوزیس بر مسیرهای التهابی و مرگ سلولی در رده سلولی A549 اپیتلیال ریه Investigation of the impact of drug-sensitive and drug-resistant Mycobacterium tuberculosis Strains on inflammatory pathways and cell death in A549 lung epithelial cells ميكروبيولوژي Microbiology تجربي Experimental سابقه و هدف: سل یکی از چالش‌های مهم سیستم بهداشتی است و تعامل میزبان با باکتری مایکوباکتریوم توبرکلوزیس در پیشرفت عفونت نقش کلیدی دارد. این عفونت از طریق تأثیر بر فرایندهای آپوپتوز، نکروز و التهاب، به رشد درون‌سلولی باکتری کمک می‌کند. اما ارتباط دقیق میان این مسیرها به‌طور کامل شناخته نشده است. روش بررسی: سویه‌های حساس و مقاوم مایکوباکتریوم توبرکلوزیس از انستیتو پاستور ایران جمع‌آوری شده و رده سلولی A549 آلوده شدند. بیان ژن‌های دخیل در مسیرهای آپوپتوز، نکروز و التهاب با استفاده از RT-PCR بررسی شد.  یافتهها: در سویه‌های حساس و مقاوم، افزایش بیان ژن‌های ضد آپوپتوزی (BCL2، RB1)، کموکاین‌ها (IL8، MCP1) و سیتوکین‌های پیش‌التهابی (TNF-α، IFN-γ) مشاهده شد، درحالی‌که بیان ژن‌های ضدالتهابی (IL10) و پیش‌آپوپتوزی (BAD، BAX) کاهش یافت. همچنین، ژنوتیپ Beijing (سویه حساس) نسبت به ژنوتیپ New-1 (سویه‌های مقاوم) بیان بیشتری در ژن‌های التهابی و آپوپتوزی داشت. هیچ تغییر معناداری در بیان ژن‌های نکروز (FADD و RIPK1) در هیچ‌کدام از سویه‌ها مشاهده نشد. نتیجهگیری: تفاوت در بیان ژن‌های مرتبط با آپوپتوز و التهاب بیشتر به ژنوتیپ‌ها وابسته بود تا مقاومت دارویی. این یافته‌ها نشان می‌دهد که ژنوتیپ باکتری نقش مهمی در شدت پاسخ التهابی و آپوپتوز در عفونت سل ایفا می‌کند.   Background: Tuberculosis has long been a significant challenge to global healthcare. The host-pathogen interaction with Mycobacterium tuberculosis plays a critical role in the progression of the infection, influencing immune responses and regulating cell death processes such as apoptosis and necrosis. However, the relationship between these processes and inflammation remains unclear. This study examines the expression levels of genes related to apoptosis, necrosis, and inflammation in A549 lung epithelial cells infected with drug-sensitive and drug-resistant Mycobacterium tuberculosis strains. Materials and methods: Strains of Mycobacterium tuberculosis with varying antibiotic resistance (sensitive, rifampin-resistant, multidrug-resistant, and extensively drug-resistant) were obtained from the Pasteur Institute of Iran. A549 cells were cultured and infected. Genes linked to the studied pathways were selected using bioinformatics, and their expression levels were assessed using RT-PCR. Results: All strains showed increased expression of anti-apoptotic genes (BCL2, RB1), chemokines (IL8, MCP1), and pro-inflammatory cytokines (TNF-α, IFN-γ), alongside reduced expression of anti-inflammatory genes (IL10) and pro-apoptotic genes (BAD, BAX) post-infection. The Beijing genotype (sensitive strain) induced higher inflammatory and apoptotic gene expression than the New-1 genotype (resistant strains). No significant changes were observed in necrosis-related genes (FADD, RIPK1). Conclusion: Variations in apoptosis and inflammation-related gene expression appear to stem more from genotypic differences than drug resistance.
