fa تاثیر فکالی‌باکتریوم بر الگوی میکروبیوتای دستگاه گوارش و پاسخ‌های التهابی در مدل موش بزرگ آزمایشگاهی وابسته به فنتانیل The effect of Faecalibacterium on the gastrointestinal microbiota pattern and inflammatory responses in a fentanyl-dependent rat model ميكروبيولوژي Microbiology كارآزمايي باليني Controlled/Randomized clinical trial <v:line from="3.6pt,2.8pt" id="_x0000_s1026" strokeweight="1.5pt" style="position:absolute; left:0; text-align:left; z-index:251658240" to="480.6pt,2.8pt"> <w:wrap anchorx="page"> </w:wrap></v:line><img alt="" src="file:///C:/Users/afsane/AppData/Local/Temp/msohtmlclip1/01/clip_image001.gif" style="width:639px; height:2px" >چکیده سابقه و هدف: بحران فعلی سلامت عمومی ناشی از اختلال مصرف اپیوئید با افزایش شیوع مصرف اپیوئیدهای سنتتیک، به ویژه فنتانیل، تشدید شده است، که منجر به افزایش موارد اوردوز می‌شود. برای کاهش تأثیرات مضر سوء‌مصرف فنتانیل، شناسایی نشانگرهای بیولوژیک جدید و راهبردهای درمانی ضروری است. روش بررسی: مطالعه حاضر به بررسی اثر فکالی‌باکتریوم بر کنترل عواقب ناشی از سوء‌مصرف فنتانیل در ترکیب جمعیت میکروبیوتای دستگاه گوارش و پاسخ‌های التهابی مرتبط در یک مدل موش صحرایی با استفاده از تکنیک‌هایReal-time PCR  مطلق و نسبی پرداخت.  یافتهها: در گروه تیمار شده با فنتانیل، فراوانی کلستریدیوم (69/0P=) و باکتروئیدس (04/0P=) افزایش و فراوانی فکالی‌باکتریوم (149/0P=) و لاکتوباسیلوس (44/0P=) کاهش یافت. گاواژ فکالی‌باکتریوم باعث افزایش معنیدار فراوانی فکالی‌باکتریوم (0074/0P=) و لاکتوباسیلوس (165/0P=) و کاهش فراوانی کلستریدیوم (166/0P=) و باکتروئیدس (013/0P=) شد. سطوح بالای سیتوکین‌های التهابی دستگاه گوارش، مانند IL-1β (01/0P=)،TNF-α  (083/0P=) و IL-6 (17/0P=) در گروه وابسته به فنتانیل، وجود التهاب دستگاه گوارش را در پاسخ به قرارگیری در معرض اپیوئید تأیید کرد. کنترل التهاب دستگاه گوارش درنتیجه گاواژ فکالی‌باکتریوم مشاهده شد (0004/0P=). نتیجهگیری: این مطالعه رابطه بین سوء‌مصرف فنتانیل، باکتری‌های دستگاه گوارش و التهاب را نشان داد. تغییرات در گونه‌های خاص باکتری ممکن است به افزایش التهاب دستگاه گوارش منجر شود و با اصلاح جمعیت میکروبیوتای دستگاه گوارش می‌توان استراتژی‌های کارآمدی را برای کنترل موثرتر اثرات ناشی از سوء‌مصرف فنتانیل و همچنین سایر اوپیوئیدها را پیشنهاد کرد.   Background: The current public health crisis resulting from opioid use disorder has been exacerbated by the increasing prevalence of synthetic opioids, particularly fentanyl, leading to a rise in overdose cases. To mitigate the harmful effects of fentanyl abuse, the identification of new biological markers and therapeutic strategies is essential. Materials and methods: The present study investigated the effect of fecal bacteria on controlling the consequences of fentanyl abuse in the composition of the gastrointestinal microbiota and related inflammatory responses in a rat model using absolute and relative Real-time PCR techniques. Results: In the fentanyl-treated group, the frequency of Clostridium (P=0.69) and Bacteroides (P=0.04) increased, while the frequency of Faecalibacterium (P=0.149) and Lactobacillus (P=0.44) decreased. Faecalibacterium gavage significantly increased the frequency of Faecalibacterium (P=0.0074) and Lactobacillus (P=0.165), while reducing the frequency of Clostridium (P=0.166) and Bacteroides (P=0.013). Elevated levels of gastrointestinal inflammatory cytokines, such as IL-1β (P=0.01), TNF-α (P=0.083), and IL-6 (P=0.17) in the fentanyl-dependent group, confirm the presence of gastrointestinal inflammation in response to opioid exposure. Control of gastrointestinal inflammation was observed as a result of Faecalibacterium gavage (P=0.0004). Conclusion: This study demonstrated the relationship between fentanyl abuse, gut bacteria, and inflammation.
