fa مقایسه اثرات عصاره هیدرو‌الکلی هندوانه‌ ابوجهل و داروی پیوگلیتازون بر انسولین، گلوکز سرم، HbA1c، رزیستین، TNF-α و ظرفیت آنتی اکسیدانی تام (TAC) در رت‌های نر دیابتی نوع II Comparative study of the effects of hydroalcoholic extract of Citrullus colocynthis and pioglitazone on Insulin, Serum Glucose, HbA1c, Resistin, TNF-α, and Total Antioxidant Capacity (TAC) in type 2 diabetic male rats گوارش Gastrology نيمه تجربي Quazi-Experimental سابقه و هدف: دیابت نوع ۲ یکی از شایع‌ترین اختلالات متابولیک جهان است که تعداد مبتلایان آن رو به افزایش است. داروهای رایج و تزریق انسولین در کنترل آن مؤثرند، اما عوارض جانبی زیادی دارند. به همین دلیل، تحقیق درباره درمان‌های گیاهی مانند  Citrullus colocynthis به‌عنوان جایگزینی مؤثر و کم‌عارضه اهمیت یافته است. روش بررسی: در این مطالعه، 60 راس رت صحرایی نر در 6 گروه دهتایی قرار داده شدند. سپس رتها با استفاده از استرپتوزوتوسین دیابتی شدند (به جز گروه شاهد). پس از تایید دیابتی شدن و اندازهگیری قندخون، حیوانات به مدت سه هفته متوالی با دوزهای mg/kg100، mg/kg 200، mg/kg 400،  عصاره هیدروالکلی و mg/kg10 پیوگلیتازون تیمار شدند. سپس انسولین، گلوکز سرم، HbA1c، رزیستین، TNF-α و ظرفیت آنتی اکسیدانی تام (TAC) اندازهگیری شدند. در پایان اطلاعات جمعآوری شده با استفاده از نرم افزار SPSS تحلیل شدند.  یافته ها: پس از القای دیابت با استرپتوزوتوسین، میانگین گلوکز خون در گروه‌های دیابتی نسبت به گروه کنترل به‌طور معنی‌داری افزایش یافت (001/0p<). درمان با عصاره هیدروالکلی Citrullus colocynthis و پیوگلیتازون موجب کاهش معنی‌دار گلوکز خون و HbA1c نسبت به گروه دیابتی شاهد شد. همچنین سطح رزیستین و TNF-α در گروه دیابتی شاهد افزایش معنی‌داری داشت و تیمار با عصاره و پیوگلیتازون باعث کاهش این شاخص‌ها گردید. از سوی دیگر، سطح انسولین و ظرفیت آنتی‌اکسیدانی تام (TAC) در گروه دیابتی شاهد کاهش یافته بود، در حالی‌که درمان با عصاره و پیوگلیتازون موجب افزایش معنی‌دار آن‌ها شد. نتیجهگیری: در مجموع به نظر میرسد که عصاره هندوانه ابوجهل با افزایش انسولین، کاهش قند خون، کاهش میزان هموگلوبین گلیکوزیله، کاهش آدیپوکاین مستعدکننده فرد به دیابت نوع 2  یعنی Resistin ، کاهش فاکتورهای التهابی (TNF-α) و افزایش ظرفیت آنتی اکسیدانی تام ، درمان موثری جهت کنترل دیابت و پیشگیری از عوارض ناشی از آن، مانند اختلالات قلبی-عروقی باشد.   Background: Type 2 diabetes is one of the most common metabolic disorders worldwide, and the number of patients is rapidly increasing. Although conventional drugs and insulin injections help to control it, they often are associated with side effects. Therefore, research on herbal treatments such as Citrullus colocynthis has gained importance as an effective and safer alternative. Materials and methods: In this study, 60 male rats were studied in 6 groups of ten, then the rats were made diabetic using streptozotocin (except the control group). After confirming diabetes and measuring blood sugar, the animals were treated with doses of 100 mg/kg, 200 mg/kg, 400 mg/kg, hydroalcoholic extract and 10 mg/kg pioglitazone for three consecutive weeks; then insulin, serum glucose, HbA1c, resistin, TNF-α and total antioxidant capacity (TAC) were measured. Finally, the collected data were analyzed using SPSS software. Results: After induction of diabetes with streptozotocin, the mean blood glucose level in diabetic groups significantly increased compared with the control group (p<0.001). Treatment with the hydroalcoholic extract of Citrullus colocynthis and pioglitazone significantly reduced blood glucose and HbA1c levels compared with the diabetic control group. In addition, resistin and TNF‑α levels were significantly elevated in the diabetic control group, while treatment with the extract and pioglitazone significantly decreased these markers. Furthermore, insulin levels and total antioxidant capacity (TAC) were reduced in the diabetic control group, whereas treatment with the extract and pioglitazone significantly increased these parameters. Conclusion: Overall, it seems that the extract of Citrullus colocynthis is an effective treatment for controlling diabetes and preventing its complications, including cardiovascular disorders, by increasing insulin, reducing blood sugar, reducing glycosylated hemoglobin, reducing the adipokine that predisposes an individual to type 2 diabetes, namely Resistin, reducing inflammatory factors (TNF-α) and increasing total antioxidant capacity.   هندوانه‌ابوجهل, پیوگلیتازون, دیابت نوع 2, گلوکز خون,رزیستین, انسولین, فاکتور نکروز تومور, HbA1c, TAC.
