نقش و اهمیت متیلاسیون DNA در روند اسپرماتوژنز

سابقه و هدف: متیلاسیون DNA یکی از عوامل تنظیم اپی‌ژنتیکی است که به صورت de novo ایجاد شده، سپس تثبیت، و از طریق تقسیم سلولی در نسل های متوالی حفظ می‌شود. این فرآیند توسط آنزیم‌های DNA متیل ترانسفراز کاتالیز می‌شود. متیلاسیون DNA در رده­ی زایا به‌اندازه­ی ایجاد متیلاسیون در گامت­ها اهمیت دارد. از آنجایی که در تنظیم بیان ژن در نسل بعدی نقش دارد و عدم ایجاد الگوهای درست متیلاسیون DNA در رده‌های زایا عواقب جدی برای تکوین پس از لقاح دارد. اختلال در تنظیمات اپی‌ژنتیکی، به‌ویژه متیلاسیون DNA اسپرم، نقش مهمی در ایجاد بیماری‌های متعدد و تأثیرات منفی بر باروری مردان داشته باشد. هدف این مطالعه مروری بررسی متیلاسیون DNA و آنزیم‌های دخیل در این فرایند و همینطور عوامل محیطی طی فرایند اسپرماتوژنز و لقاح بود.
روش بررسی: جستجو در بانک‌های اطلاعاتی نظیر Scopus، Pubmed و Google Scholar براساس کلید واژه‌ها انجام گرفت. سپس مقالاتی که دارای معیار ورود به مطالعه بودند بین سال‌های 1975 تا 2017 بررسی شدند. 
یافته­ ها: اختلالات بیان آنزیم‌هایDNA methyltransferase استرس اکسیداتیو سلولی را افزایش می‌دهد و سبک زندگی نیز می‌تواند الگوهای متیلاسیون DNA طی اسپرماتوژنز و پتانسیل باروری مردان را تحت تاثیر قرار دهد.
نتیجه ­گیری: از آنجایی که متیلاسیون DNA اسپرم ارتباط نزدیکی با ناباروری مردان دارد، درک مکانیسم‌های زیر بنایی متیلاسیون DNA به‌منظور توسعه استراتژی‌های درمانی مهم است.
