fa بررسی خواص ضدجهشی و سایتوتوکسیک عصاره پوست سیب Malus domestica زيست شناسي مولكولي Molecular Biology تجربي Experimental <p dir="RTL">سابقه و هدف<em>:</em> <em>در حال حاضر سرطان یکی از ع</em><em>ل</em><em>ل </em><em>عمده </em><em>مرگ و می</em><em>ر</em><em> در جهان است</em><em> بنابراین از پیشگیری می&shy;توان به عنوان مهم&shy;ترین رویکرد درمانی سرطان درنظر گرفت. رژیم غذایی نقش مهمی در پیشگیری از سرطان بازی می&shy;کند. امروزه محققان توجه ویژه&shy;ای به غذاهای طبیعی دارند که می&shy;توانند به طور چشمگیری نقش پیشگیری&nbsp; از&nbsp; سرطان را داشته باشند. </em><em>هدف </em><em>از </em><em>این تحقیق سنجش</em> <em>خواص ضدجهشی و ضدسرطانی عصاره پوست سیب بود.</em></p> <p dir="RTL">روش بررسی<em>:</em> <em>در این تحقیق تجربی، سلول&shy;های سرطانی سندروم سزاری یا&nbsp; لنفوم </em><em><span dir="LTR">T</span></em><em> انسانی </em><em><span dir="LTR">(HUT-78)</span></em> <em>در محیط </em><em><span dir="LTR">&nbsp;[Sigma] RPMI</span></em><em> کشت شدند. در این محیط کشت، </em><em>۱۰% سرم جنین گاوی </em><em><span dir="LTR">FBS(Fetal Bovine Serum)</span></em><em>، </em><em><span dir="LTR">L</span></em> <em>&ndash;</em><em>گلوتامین، پنی سیلین، و استروپتومایسین اضافه شد و در دمای 37 درجه به مدت 2 روز انکوبه گردید. سپس با عصاره پوست سیب تیمار شدند و با روش </em><em><span dir="LTR">MTT</span></em><em>، توان زیستی سلول&shy;ها بررسی شد. همچنین خواص ضدجهشی عصاره پوست سیب بر حسب روش استاندارد سنجش جهش برگشتی&nbsp; (آزمون ایمز) مورد بررسی قرار گرفت. </em></p> <p dir="RTL">یافته&shy;ها: <em>در روش </em><em><span dir="LTR">MTT</span></em><em>،</em><em> سلول&shy;های سرطانی لنفوم </em><em><span dir="LTR">T</span></em><em> انسانی مرگ سلولی معنی&shy;داری را نسبت به گروه&shy;های شاهد نشان دادند (01/0</em><em><span dir="LTR">p<</span></em><em>)</em><em>. در آزمون ایمز، عصاره پوست سیب از موتاسیون برگشتی جلوگیری نمود و درصد بازدارندگی در سنجش ضدجهشی 3/81 درصد بود. </em></p> <p dir="RTL"><strong>نتیجه</strong>&shy;<strong>گیری</strong><em>:</em> <em>در این بررسی، خواص ضدسرطانی عصاره پوست سیب بر روی سلول های سرطانی در شرایط </em><em><span dir="LTR">in vitro</span></em> &nbsp;<em>مشخص گردید و می&shy;تواند گامی بر ترویج فرهنگ استفاده از پوست سیب در رژیم غذایی جامعه باشد</em><em>.</em></p> <p dir="RTL"><strong>واژگان کلیدی:</strong> <em>ضدجهشی، ضدسرطانی، پوست سیب، سلول</em><em>&shy;</em><em>های سرطانی لنفوم </em><em><span dir="LTR">T</span></em><em> انسانی، آزمون ایمز.</em></p> <p><strong>Background</strong>: At present, cancer is one of the leading causes of mortality in the world. Therefore, prevention can be considered as important as treatment in the management of cancer. Diet can play a vital role in cancer prevention. Nowadays, scientists are looking for natural food which can prevent the cancer occurrence. The purpose of this research was to examine antimutagenicity and anticancer effects of apple peel extract.</p> <p><strong>Materials and methods</strong>: In this experimental study, HUT-78 cell line were cultured in 90% RPMI1640, supplemented with 10% fetal calf serum, l- glutamine, peniciline, streptomycin and then incubated at 37^&deg;c for 2 days. The cancer cell line was treated by different apple peel extract concentrations and cellular vital capacity was determined by MTT assay. The apple peel extract was subsequently evaluated in terms of antimutagenicity and anticancer properties by a standard reverse mutation assay (Ames test).</p> <p><strong>Results</strong>: During MTT assay, T cell lymphoma cancerous cells revealed significant cell death compared with controls (p<0.01). In Ames test, the apple peel extract prevented the reverted mutations and the hindrance percent was 81.3%.</p> <p><strong>Conclusion</strong>: The results demonstrated that apple peel extract has anticancer effects in in vitro condition.
