مروری بر کاربرد CRISPR-CAS در بیوتکنولوژی میکروبی

امروزه استفاده از سیستم‌های CRISPR-Cas برای ویرایش و اصلاح ژنوم در گونه‌های مختلف میکروبی به طور گسترده مورد استفاده قرار می­گیرند. سیستم‌های CRISPR-Cas که ابتدا به‌عنوان مکانیسم دفاعی باکتری‌ها در برابر عوامل مهاجم ویروسی شناسایی شدند، در طیف گسترده‌ای از گونه‌های میکروبی، از جمله باکتری‌ها و آرکئا، وجود دارند. این سیستم‌ها به دو کلاس اصلی تقسیم می‌شوند: کلاس ۱ که شامل مجموعه‌های پروتئینی چندزیرواحدی است، و کلاس ۲ که از طریق سیستم‌های پروتئینCas  تک واحدی مانند (Cas9) متمایز می‌شوند. انعطاف‌پذیری سیستم‌های CRISPR-Cas یکی از جذاب‌ترین ویژگی‌های آنها است. روشهای مختلف ویرایش ژنوم در سیستم‌های کلاس ۱ و ۲ نشان از انعطاف‌پذیری بالای این سیستم‌ها در تنوع مکانیسم‌های دفاعی آنها، که منجر به محافظت در برابر عفونت‌های ویروسی می شوند. علاوه بر آن سیستم‌های CRISPR-Cas در پایداری و یکپارچگی ژنوم باکتری‌ها و آرکئا نقش دارند. این سیستم‌ها اکنون ابزارهای ضروری در آزمایشگاه برای ژنومیک کارکردی، ویرایش دقیق ژنوم و برنامه‌ریزی هدفمند ژن هستند. آنها نقش کلیدی در زیست‌فناوری دارند و در مهندسی میکروارگانیسم‌ها و استفاده از آنها در تصفیه محیط‌زیست و تولید سوخت زیستی نقش ایفا می‌کنند. این سیستم‌ها علاوه بر آزمایشگاه در عرصه پزشکی نیز کاربرد دارند. تشخیص‌های مبتنی بر CRISPR امکان شناسایی سریع و دقیق عفونت‌ها و تغییرات ژنتیکی را فراهم آورده و به تشخیص زودهنگام بیماری‌ها کمک می‌کند. علاوه بر این، توسعه راهکارهای هدفمند ضدمیکروبی با استفاده از  CRISPRنیز برای مقابله با میکروارگانیسم‌های مقاوم به دارو کاربرد وسیعی یافته است. در مجموع، سیستم‌های CRISPR-Cas به ابزاری کلیدی در مهندسی ژنوم میکروارگانیسم‌ها و توسعه راهکارهای نوین تشخیصی و درمانی در پزشکی تبدیل شده‌اند. در این مقاله مروری، داده‌ها با جست‌وجو در پایگاه‌های Springer، ScienceDirect، Scopus وJohn Wiley  طی سال‌های ۲۰۱۸ تا ۲۰۲۵ و بر اساس واژه‌های کلیدی تعیین‌شده در سایت MeSH گردآوری و تحلیل شدند.
