اثر ورزش تردمیل کوتاه‌مدت بر بیان ژن‌های PINK-1, Parkin و PGC-1α در قلب موش‌های حساس‌شده به نیکوتین

چکیده:   (1184 مشاهده)
سابقه و هدف: نیکوتین در قلب بیان ژن­های مختلفی را تحت تاثیر قرار می­دهد. پروتئین PINK-1 (PTEN-induced kinase 1) تنظیم­کننده اصلی میتوفاژی سلولی است. از طرف دیگر، Parkin پروتئینی است که در روند یوبیکیتیناسیون نقشی اساسی دارد و باعث القای مسیرهای نجات سلولی می­گردد. همچنین، PGC-1α (Peroxisome proliferator-activated receptor gamma coactivator 1-alpha) تنظیم­کننده اصلی بیوژنز میتوکندری است. از سوی دیگر، ورزش اثرات مثبت فیزیولوژیک بسیاری بر بیمارانی دارد که از نارسایی قلبی رنج می­برند. بنابراین هدف از این پژوهش  تاثیر تمرین ورزش  کوتاه مدت  استقامتی بر بیان ژن­های Parkin، PINK-1 و PGC-1α در قلب موش­های حساس­شده به نیکوتین بود.
روش بررسی: در این پژوهش از موش­های نر نژاد ویستار، با وزن تقریبی 180 تا 200 گرم استفاده شد. حیوان­ها  نیکوتین  را در مقدار 21/0 میلی­گرم به ازای هر کیلوگرم وزن موش به صورت درون صفاقی دریافت کردند. برای بررسی بیان ژن­های مورد مطالعه، در هر گروه بررسی بافت­ها با تکنیکPCR  Real Time انجام شد. 
یافته­ها: نتایج نشان داد که نیکوتین بیان ژن  PGC-1αرا کاهش داد (05/0P<) و بر دیگر ژن­ها تاثیری نداشت (05/0P>). همچنین، ورزش کوتاه مدت استقامتی بیان تمام ژن­ها را تا حدی کم افزایش داد که از نظر آماری معنی­دار نبود (05/0P>).
نتیجه­گیری: به­نظر می­رسد ورزش کوتاه مدت استقامتی می­تواند تاثیرات پرو-آپوپتوتیک و تحریک­کننده­ی استرس اکسیداتیو ناشی از نیکوتین را کاهش دهد. علاوه بر این، تمرین ورزشی طولانی­ مدت احتمالا بتواند بر ژن­های موثر بر میتوفاژی تاثیری معنی­دار و مثبت بگذارد.
